Upload screening data

  1. Before uploading your screening data (primary or CRC), please ensure that you have entered your assay target (HTS Screens > Targets), method (HTS Screens > Methods), and controls (HTS Screens > Methods > Assay Details > Assay Controls) in RxPlora.
  2. Select HTS Screens > Primary Screens > Import data for uploading primary screening data and HTS Screens > CRC Screens > Import data for uploading CRC data. Enter an assay plate template for your assay by going to HTS Screens > Assay Plate Templates. This saves and re-uses control well positions for future plate uploads. You will select the assay plate template in the second step of the upload process.

    • To upload Plate Reader files, select the appropriate options for your data file(s), then use the ‘Choose File’ button to select the .zip file containing your assay plate data. After uploading, select your desired ‘PARTS’ from the plates, and enter the assay barcode and stock plate number for the first plate (or upload a mapping file). Once the csv file is generated, you can choose to save it for your records. Then click ‘Upload file’.
    • To upload an RxPlora compatible file, click the ‘Upload RxPlora compatible file’ tab underneath the ‘Upload’ button. Verify that your data is formatted in a comma-separated value (csv) file according to the template specified in the sample file provided, then click the ‘Choose File’ button to upload the formatted data file.
  3. First, verify your data in the upload task. RxPlora parses the formatted data file you uploaded and asks you to select the assay target, method, controls, and additional measurements, if applicable. If you entered your target, method, and controls in Step 1, they should automatically appear in the selection drop-down. Once you select all the correct parameters, then click ‘Create Assay’.
  4. Second, define the assay plate layout. If you have a saved assay plate template, select that from the assay plate template drop-down. If not, use the selection radio buttons to select the wells that contain the controls. Then click the ‘View Results’ button.

  5. RxPlora calculates the percents, standard deviations, and other relevant assay parameters based on the control selection in the previous step and takes you to the assay plate view for the uploaded assay.
